Nogalonic acid methyl ester cyclase, a polyketide cyclase in the biosynthetic pathway of the antibiotic nogalamycin, has been crystallized. The crystals obtained in the presence of the substrate are orthorhombic, space group I222, with unit-cell parameters a = 69.1, b = 72.0, c = 65.4 Å and diffract to 1.35 Å resolution.

The structures of three closely related NTF2-like superfamily proteins that are involved in type II polyketide biosynthesis in as-yet-unknown ways were determined. The presence of a solvent-accessible cavity and the conservation of the His/Asp dyad suggest a potential enzymatic role for these enzymes in polyketide biosynthesis.

The cofactor-independent monooxygenase SnoaB from the nogalamycin-biosynthesis pathway and a designed selenomethionine-substituted double mutant were crystallized in space group P21212. The native enzyme diffracted to a resolution of 2.4 Å.

The crystal structure of IdmH from the biosynthetic gene cluster for indanomycin is presented. NMR data show that this enzyme binds its postulated product, indanomycin, and QM/MM modelling of the reaction strongly supports the view that IdmH catalyses indane-ring formation in indanomycin biosynthesis via a Diels–Alder reaction.
