A novel three-dimensional X-ray microtomographic micro-imaging system which enables simultaneous measurement of differential phase contrast and absorption contrast has been developed. The optical system consists of a scanning microscope with one-dimensional focusing device and an imaging microscope with one-dimensional objective.

A full-field X-ray diffraction contrast method is presented (high-resolution 3D X-ray diffraction), for 3D mapping of plastically deformed microstructures. The essence of the method is the introduction of a 2D detector in the optical mid-field regime. The properties and limitations of the method are estimated by numerical simulations.

The application of X-ray coherent diffractive imaging to structure determination of single biomolecules at free electron lasers yields signal levels down to a few hundred scattered photons per frame or less. Here we demonstrate in an analog experiment at a synchrotron source that even at a sparsity level of order 1.3 × 10−3 photons per pixel per frame the 3D structure of a nano-fabricated sample can be obtained, even if the orientation of the particle for a given individual frame is not explicitly known.

A microcrystal-prelocation method is demonstrated using low-dose 2D full-field propagation-based X-ray phase-contrast imaging on samples with an essentially flat geometry for automated serial crystallography data collection at a microfocus macromolecular crystallography beamline.
