The synthesis of nonribosomal peptides often introduces D-configured amino acids into the final products. The first crystal structure of the epimerization domain of a nonribosomal peptide synthetase reveals a bilobal architecture and implies a general acid–base mechanism for the LD isomerization of aminoacyl substrates.
